Sample Collection (Nasopharyngeal):
Gently tilt the patient’s head back 45°-70° to straighten the nasal passage. Gently rub the surface with the swab, rotating it 3-4 times. Leave the swab in place for a few seconds to absorb secretions. Slowly remove the swab, rotating it.
Sample Collection (Oropharyngeal):
Collect the sample before eating or 2-3 hours after a meal. The patient should rinse their mouth with warm boiled water beforehand. Gently insert the swab between the tonsils and uvula. Move the swab back and forth to collect material from the back of the throat, tonsils, and any inflamed or ulcerated areas. Avoid touching the cheeks, tongue, gums, or collecting saliva.
Test Procedure:
Place the swab in the vial containing the buffer solution. Rinse the swab by rotating it against the vial walls at least 10 times. Squeeze the swab to extract the remaining fluid. Close the vial and discard the swab. Shake the vial vigorously. Break off (or cut) the tip of the dropper. Add 3 drops to the test cassette’s sample well. Visually assess the results after 10 minutes.
Result Interpretation:
One red control line (C) in the test window indicates a negative result (no SARS-CoV-2 nucleocapsid antigen detected). Two parallel lines indicate a positive result (SARS-CoV-2 nucleocapsid antigen detected). The absence of a control line indicates an invalid result; repeat the test using a new RED SARS-CoV-2 Ag test cassette. False positive or negative results can occur due to expired kits, improper sample collection, or temperature fluctuations during transport, storage, and/or use.
