Dopaminergic effects of the drug, causing a decrease in prolactin production, eliminate hyperprolactinemia. An increased concentration of prolactin disrupts the secretion of gonadotropins, as a result of which disturbances in the maturation of follicles, ovulation and the formation of the corpus luteum may occur, which further leads to an imbalance between estradiol and progesterone and can cause menstrual irregularities, as well as mastodynia. Unlike estrogens and other hormones, prolactin also has a direct stimulating effect on the proliferative processes in the mammary glands, enhancing the formation of connective tissue and causing the expansion of the milk ducts. A decrease in the content of prolactin leads to the reverse development of pathological processes in the mammary glands and relieves pain. The rhythmic production and normalization of the ratio of gonadotropic hormones leads to the normalization of the second phase of the menstrual cycle. Pharmacodynamic properties In the literature, it has been reported that water-alcohol extracts of the common plantain (Agnus castus) inhibit the release of prolactin in vitro. An action aimed at inhibiting the release of prolactin has also been observed in animal experiments. Conducted clinical studies have shown that the administration of an extract of the common rod (Agnus castus) to women with moderate signs of elevated prolactin concentrations or with increased release of prolactin under stress (so-called “latent hyperprolactinemia”) led to a decrease in prolactin levels. In vitro studies have shown that lactotropic pituitary cells are the site of action, with the mechanism of action being the dopaminergic principle. Bicyclic diterpenes have been identified as a group of substances that contribute to the action of the chamomile extract in reducing prolactin levels. These substances bind to the human dopamine receptor subtype 2 and dose-dependently decrease prolactin release in cultured rat pituitary cells. Pharmacokinetics No data available. Dosing and Administration The recommended dose of Prefemin is 1 tablet per day. The tablet is intended for oral administration, it should not be chewed or dissolved, but must be swallowed whole with a glass of water.
